In a previous project, a novel mutational signature associated with replication strand bias was reported in hematopoietic stem and progenitor cells isolated from hematopoietic stem cell transplanted patients. DNA replication has been shown to contribute to cancer risk and to be extremely important in the context of mutagenesis, however the exact manner in which it influences mutational mechanisms is still unknown. In eukaryotic cells, DNA replication relies on DNA polymerases, which are essential for DNA repair and translesion DNA synthesis (TLS). The aim of my project is to elucidate whether altered expression of TLS polymerases can cause this previously reported mutational signature
