Conditional protein degradation system to study the role of SMARCB1 in rhabdoid tumor organoid models

Rhabdoid tumors are a type of pediatric cancer that is mainly characterized for the presence of a mutation in SMARCB1, a member of the SWI/SNF complex. My project consists in developing a system in which this gene can be re-expressed in a time-controlled manner in tumor organoid models. This on/off switch is achieved by a dTAG that degrades the re-expressed SMARCB1 protein in the organoid models. With this, we aim to study the role of SMARCB1 in the development and maintenance of the phenotypic and epigenetic landscape of rhabdoid tumor cells.
